It's a sequence from the documentary film
The Aristocrats which explores the history of that joke. It's as old as stand-up comedy pretty much, and has been told by virtually every comedian during their career at some point. The punchline is always the same and the challenge is to come up with - usually just as an improv - the longest, most vulgar string of things you can imagine for that family to do to one another.
That's Trey Parker's version, Bob Saget's is just wicked, and one of my favorites comes from Richard Jeni:
